London Buses route 427 is a Transport for London contracted bus route in London, England. Running between Uxbridge and Acton, it is operated by Metroline.

Route 427 commenced operating on 9 April 2005 operating between Acton Market Place and Uxbridge Station via Ealing - Hanwell - Southall - Hayes End - Hillingdon Hill. The route was initially operated by First London from their Acton (AT) garage using Plaxton President bodied Dennis Tridents. The route was introduced to replace the withdrawn section of route 207 between Hayes By Pass and Uxbridge.

Transport for London were proposing to build the West London Tram between Shepherd's Bush and Uxbridge, running along Uxbridge Road. The tram would have replaced the current routes 207, 427 and 607. Transport for London claimed that trams would carry more passengers and be more reliable, but due to the Crossrail project going ahead the plans were scrapped.

On 10 April 2010, the route was retained by First London with brand new Wright Eclipse Gemini 2 bodied Volvo B9TLs introduced.

On 21 February 2011, the route was extended in Acton from Horn Lane to Town Hall.

On 22 June 2013, the route was included in the sale of First London's Hayes garage (HS) to Metroline.

Abellio London will take over the operation of route 427 operating from their Hayes (WS) garage with brand new Enviro 400 MMC bodied Alexander Dennis E40Hs introduced from 8 April 2017.
